It’s the first time we’ve seen him see through a Loki illusion, the first time he’s “not fall[en] for that”, as Loki put it so mockingly in The Avengers. And I love the look on Loki’s face before he drops the illusion. That, “Why now, of all times?!” moment of shock.
I don’t want to give Thor too much credit or Loki too little, but I do think that Loki’s previous illusions were both in extremely tense situations, where Thor couldn’t afford to risk being wrong. His brother hanging off the edge of the Bifrost, asking for help; his brother about to escape on the helicarrier and wreak havoc… they required quick decisive action, and especially in the case of the Bifrost I think it was worth the chance that it was a trick to potentially save Loki from falling.
Here, Thor has plenty of space and plenty of time to judge, and being wrong only risks more mockery.
